摘要
为了解在新疆奶牛中是否存在牛病毒性腹泻病毒(BVDV)基因2型的感染,利用BVDV 5′-UTR特异性引物对沙湾县某牛场采集疑似感染BVDV病料样品进行RT-PCR检测,并将检测为阳性样品处理后接种MDBK单层细胞,结果从血液样品中成功分离出1株可产生细胞病变(CPE)的BVDV毒株(命名为BVDV-SW)。通过对该分离株5′-UTR和E2基因的扩增、测序和序列比较,证实所分离的毒株为BVDV基因2型。遗传进化分析表明,该分离株与17237毒株具有较近的亲缘关系。证实新疆奶牛中存在BVDV-2的感染,为该病的免疫防控提供了科学依据。
In order to know whether the cows in Xinjiang province have been infected with BVDV type 2 (BVDV-2). The suspected BVDV samples from a cattle farm in Shawan country were detected by RT-PCR using BVDV 5'-UTR specific primer. The processed positive samples were inoculated on MDBK cells monolayers. One BVDV strain was successfully isolated from the blood samples which can produce a cyto-pathic effect (CPE), and the isolated strain was confirmed to be BVDV type 2 (named BVDV-SW) by the amplification of 5' UTR and E2 gene, sequencing and sequence comparison. The phylogenetic analysis showed that the isolated strain and 17237 strain have a close genetic relationship. These results demonstra-ted that the cows in Xinjiang provincewere infected with BVDV type 2. This study provides a scientific ba- sis for the vaccination.
