摘要
糖苷酶底物在病原菌检测中具有十分重要的作用,为了进一步开发新的特异性细菌检测方法,设计合成了一种新颖的基于荧光素Tokyo-Green的标题化合物。首先以廉价易得的D-半乳糖为原料,经过4步反应合成得到四乙酰D-岩藻糖;然后和荧光素Tokyo-Green进行Koenigs-Knorr糖苷化反应得到目标产物,该酶底物可以用于β-D-岩藻糖苷酶的特异性检测。
Various glycoside enzymatic substrates have been customized for detecting an expanding range of target microor-ganisms. In order to develop highly specific microbial assays, a novel enzyme substrate for β-D-fueosidase using Tokyo- Green as the fluorogenic group has been designed. Firstly,tet-raacetates D-fucose was synthesized from the commercially in- expensive D-galactose in 4 steps in moderate overall yield. Then, the corresponding 1-α-bromide-2,3,4-triacetates was coupled to fluoreseein Tokyo-Green using Koenigs-Knorr gly- cosylation to provide the target β-D-fucosidase substrate. It was an excellent enzyme substrate for β-D-fucosidase.
