高效液相色谱法测定蛇床子素微乳中蛇床子素的含量

Determination of the Osthole content in Osthole microemulsion by HPLC

目的:建立高效液相色谱法(HPLC)测定蛇床子素微乳中蛇床子素含量的方法,为蛇床子素微乳的质量控制提供依据。方法:采用Diamonsil C18(4.6mm×150mm,5μm)色谱柱,流动相甲醇-水(70∶30),检测波长322nm,流速1.0mL/min,柱温30℃。结果:蛇床子素在0.096-0.480mg/mL范围内呈良好的线性关系,r=0.9999,平均回收率为100.64%,RSD为1.65%。结论:HPLC测定蛇床子素微乳中蛇床子素含量的方法简便、准确、可靠,可用于蛇床子素微乳的质量控制。

Objective： To establish an HPLC method for determination of Osthole in Osthole microemulsion, and to provide some basis for the quality control of Osthole microemulsion. Methods： HPLC method was used with Diamonsil Ca8 column （4.6mm×150mm, 5μ m） and methanol-water （70：30） as mobile phase at flow rate of 1.0mL/min,with the detection wavelength of 322nm and the column temperature of 30℃. Results： This method had good linearity in the range of 0.096-0.480 mg/mL （r=0.9999）, and the average recovery was 100.64% with RSD of 1.65% （n=5）. Conclusion： The method of determination of the Osthole content in Osthole microemulsion by HPLC was established, which was simple, sensitive, reliable and practicable for the quality control of Osthole microemulsion.
