摘要
从纤维素酶产生菌绿色木霉TY-2出发,通过紫外诱变技术选育出1株遗传稳定性良好的高产菌株H-28,其产纤维素酶的滤纸酶活力稳定在2.67 U/g左右,较出发菌株提高58.08%。以麸皮和蔗渣为主要原料对变异株H-28进行固态发酵研究,单因素优化了培养基、培养条件和表面活性剂对菌株H-28发酵产纤维素酶的影响,最后选取影响产酶较大的4个因素:发酵时间、Mandels营养盐液、蛋白胨、吐温-80做4因素3水平正交试验。最终优化后突变菌株H-28的产酶能力最高值为6.79 U/g,是出发菌株的4.07倍。
The high cellulase-producing mutant strain H-28 was Screened from the parent strain Trichoderma viride TY-2 which treated with UV. The filter paper activity of cellulase was stabilized at 2.67 U/g and increased by 58.08% compared with the original strain.Select bran and bagasse as the main raw materials for solid state fermentation studies of mutant strain H-28.The effects of compo- sitions of culture medium, cultuconditions and surfactant on cellulase production were investigated. Four main factors (fermentation conditions) that affect cellulase production fermentation time, the Mandels" nutrition salt solution, peptone, Tween-80 were screened out,then chose the four factors andthreelevels to do orthogonal experiment. The final maximum FPA of cellulase produced by the optimized mutant strain H-28 was 6.79 U/g, which is 4.07 times the initial strain.
