摘要
目的研究胡黄连苷Ⅱ治疗大鼠脑缺血损伤的最佳剂量和时间窗。方法应用双侧颈内动脉结扎法建立大鼠前脑缺血模型,按照正交试验设计分组,经腹腔注射胡黄连苷Ⅱ干预治疗,酶联免疫吸附法检测血清神经元特异性烯醇化酶(NSE)、神经胶质细胞标志蛋白S100B、髓鞘碱性蛋白(MBP)的含量,综合评价胡黄连苷Ⅱ在脑缺血损伤中的疗效。结果据血清NSE、S100B和MBP含量分析,胡黄连苷Ⅱ治疗脑缺血损伤的最佳治疗时机和剂量分别为脑缺血1.5 h、腹腔注射20 mg/kg。结论从治疗时间窗最大化和用药剂量最小化原则综合分析,在脑缺血损伤治疗中,在脑缺血1.5 h腹腔注射胡黄连苷Ⅱ20 mg/kg体重疗效最好。
Objective To study the optimum dose and time window of picroside II in treatment of cerebral ischemic injm^y in rats.Methods The forebrain ischemia models were established by bilateral common carotid artery occlusion (BCCAO) methods.The successful models were randomly divided into sixteen groups according to orthogonal experi- mental design and treated by injecting picroside Ⅱ intraperitonenally at different ischemic time with different dose. The concentrations of neuron-specific enolase (NSE),neuroglial marker protein S100B and myelin basic protein (MBP) in serum was determined by enzyme linked immunosorbent assay to evaluate the therapeutic effect of picroside ]] in cerebral ischemic injury.Results The best therapeutic time window and dose of picroside ]] in cerebral isehemic in- jury was ischemia 1.5 h with 20 mg/kg body weight according to the concentrations of NSE,S100B and MBP in serum. Conclusion According to the principle of lowest therapeutic dose with longest time window,the optimized therapeutic dose and time window is injecting picroside Ⅱ intraperitonenally with 20 mg/kg body weight at ischemia 1.5 h in cerebral ischemic injury in rats.
