固定时间对乳腺癌快速冰冻切片免疫组化结果的影响

目的分析固定时间对乳腺癌冰冻切片免疫组化结果的影响，探寻最佳的固定时间。方法利用AAF固定液（配方为丙酮85mL、冰醋酸5mL、40％甲醛10mL）以1min、2min、4min、6min、8min、10min、12min、15min不同时间对乳腺癌组织冰冻切片进行固定，然后利用6种乳腺癌相应抗体进行免疫组化染色，根据2004—2008年全国十次免疫组化质控活动所用的免疫组化质控评分标准对图像质量进行评分量化。结果固定时间可以直接影响乳腺癌冰冻切片免疫组化图像的质量，最佳的固定时间为6～8min。结论对乳腺癌冰冻切片固定6～8min可取得质量最高的免疫组化结果。

Objective To evaluate the influence of fixation time on and to search for the most suitable fixation time for immunohistochemical staining of frozen sections. Methods Breast cancer＇s fast frozen sections were fixed in AAF（containing 85% acetone, 5% glacial acetic acid and 4% formaldehyde） for 1 min, 2 rain, 4 min, 6 min, 8 rain, 10 min, 12 rain and 15 rain respectly. Then Immunohistochemical staining was done using six kinds of antibodies. The staining results were assessed and quantified based on the scoring standards that used in Chinese immunohistochemical quality control activities for ten times from 2004 to 2008. Results Different fixation times could make direct influence on the quality of immunobistochemical staining of frozen sections and the most suitable fixation time was 6-8 min. Conclusion The best quality of immunohistochemical staining of breast cancer＇s fast frozen sections was achieved when the fixation time was restricted to s 6-8 min.
