摘要
目的 :研究大鼠脑缺血再灌注后生长相关蛋白 - 4 3( GAP- 4 3)表达的变化规律及外源性神经生长因子 ( NGF)的影响。方法 :成年健康雌性 Wistar大鼠 72只 ,随机分为假手术组、自然恢复组、人工脑脊液组和 NGF治疗组。采用线栓法建立大脑中动脉缺血再灌注 ( MCAO)动物模型 ,应用免疫组织化学方法观察脑缺血再灌注后 GAP- 4 3的表达。结果 :( 1)脑缺血再灌注 6h后 ,缺血周围区 GAP- 4 3表达逐渐增高 ,第 7天达高峰 ,以后逐渐降低 ,第 2 1天仍有表达。 ( 2 )应用外源性 NGF后 ,GAP- 4 3表达较对照组有所增高 ,但无显著性差异 ( P>0 .0 5)。结论 :提示中枢神经系统损伤后 ,神经元具有再生和修复的可塑性 ,外源性 NGF对 GAP- 4
Objective: To study the exporession of growth associated protein 43(GAP 43) after middle cerebral artery occlusion/reperfusion in rats, and the influence of exogenous nerve growth factor(NGF) on GAP 43. Methods: Seventy two adult female rats were randomly divided into 4 groups: sham operated group, natural recovering group, ACSF group and NGF group. The rats received left milddle cerebral artery occlusion for 2 hours. The sections of the brains were subjected to 6 hours and 1,3, 7, 14, 21 days of reperfusion, and were processed by immunochemistry with antibodies against GAP 43. Results: In the sham operated group, GAP 43 immunoreactivity was present in cortical and hippocamparal neurons. In other group: (1)GAP 43 immunoreactivity increased at 6 hours reperfusion, attained the peak at the 7th days in the boundary zone to the ischemic core, and decreased in the later days, it remain abouve the normal at 21th day. (2)The expression of GAP 43 was slightly increased after NGF treatment, but the difference was not remarkedy. Conclusion: It is suggested that there might be neuroplasticity of regeneration and repairment in central nervous system after injury. The regulative effect of expgenous NGF on GAP 43 need further study.
