黑曲霉风化含钾矿石过程中碳酸酐酶和半胱氨酸合成酶基因表达量的差异 被引量:5

摘要 研究黑曲霉(Aspergillus niger)在含有可溶性钾(KCl)或只含有非可溶性钾(含钾硅酸盐矿粉)两种条件下风化含钾硅酸盐矿粉时碳酸酐酶(CA)和半胱氨酸合成酶(CysM)的基因表达差异.采用修改的Czapek培养基(用Na2HPO4替代K2HPO4)培养黑曲霉,设置A组(添加含钾矿粉不加KCl)和B组(加KCl不加含钾矿粉)两种处理方式.分别从A组和B组两种处理培养的黑曲霉菌丝体中提取mRNA,反转录后通过实时荧光定量PCR(RT-qPCR)对CA和CysM基因的转录表达进行相对定量.构建目的基因(CA和CysM)与内参基因(甘油醛-3-磷酸脱氢酶,GAPDH)的相对标准曲线,验证目的基因和内参基因扩增效率一致,从而能对目的基因表达进行相对定量.结果表明,A组条件下该菌的CA和CysM基因表达分别比B组条件下相应的基因表达上调了1.7倍和11.7倍(即A组是B组基因表达的2.7倍和12.7倍),对与这两种酶合成相关的黑曲霉风化含钾矿物可能的代谢途径与功能进行了分析.研究结果为进一步探讨黑曲霉在风化含钾矿物过程中的代谢调控作用提供了基础资料.
