Large-scale multiplication of human umbilical cord mesenchymal stem cells in platelet lysate

摘要 目的用血小板裂解液(platelet lysate,PL)大规模扩增人脐带间充质干细胞(umbilical cord mesenchymal stem cell,UCMSC),并检测其生物学特性,为临床应用提供实验依据。方法分别采用PL和胎牛血清(FBS)低密度扩增人UCMSC,比较两组UCMSC的细胞形态、大小、克隆形成率、增殖能力、细胞表型和分化能力。结果 PL扩增的UCMSC形态细长;直径明显小于FBS扩增的UCMSC(P<0.05);克隆形成率与FBS扩增的UCMSC差异无统计学意义(P>0.05);有更高的细胞累积群倍数;与FBS扩增的UCMSC有相似的细胞表型;与FBS扩增的UCMSC均具有成骨、成脂诱导分化能力,但PL扩增的UCMSC成骨分化能力更强。结论 PL可取代FBS用于大规模扩增UCMSC。 Objective To multiply human umbilical cord mesenchymal stem cells (UCMSCs) in a large quantity in platelet lysate (PL), determine their biological property and provide an experimental basis for their clinical application. Methods Human UCMSCs were multiplied at low densities in PL-and fetal bovine serum (FBS)-supplemented media re-spectively, of which the morphology, size, clone formation rate, multiplication ability, phenotype and differentiation were compared. Results The UCMSCs multiplied in PL were thin and long, of which the diameters were significantly less than (P 〈 0. 05), while the clone formation rate showed no significant difference with (P 〉 0. 05) and the cumulative population doubling (CPD) was higher than those in FBS. The phenotype of UCMSCs in PL was similar to those in FBS. Both the UCMSCs multiplied in PL and FBS showed potentials in osteogenic and adipogenic differentiations. However, the UCMSCs were more potential in differentiation to osteocytes. Conclusion PL may be used for large-scale multiplication of UCMSCs instead of FBS.
