人子宫内膜基质细胞的体外蜕膜化及其与腺体细胞的关系 被引量:1

摘要 体外分离、培养人子宫内膜基质细胞和腺体细胞,待基质细胞生长融合后,加入孕酮刺激,使其蜕膜化。结果显示,孕酮(progesterone,P)组基质细胞胞体变大、变圆,尤以含胎牛血清(fetal bovine serum,FBS)组显著。用酶免疫分析法检测培养液中催乳素(pro-lactin,PRL)含量发现,培养d17~20PRL分泌量达到高峰;免疫细胞化学染色可见基质细胞PRL免疫反应阳性。在此基础上,将同期培养的腺细胞匀浆液加入到已蜕膜化的基质细胞中,结果发现,在加入腺体匀浆液3d后蜕膜细胞胞体变小,PRL分泌量减少,而对照组的PRL产量则递增。结果提示,子宫内膜腺细胞对蜕膜反应有一定的抑制作用,其作用方式和机制有待于进一步探讨。 The stromal cells and glands of human endometrium were isolated and cultured. After stro- mal cells had been confluent, they were induced decidualization by P stimuli. The results showed that the stromal cells in the P-treated group became larger and ovoid, especially in the FBS group. The peak PRL level in the group P was obtained at the 17th ~19th d. Immunocy- tochemical staining for PRL showed that the strong immunoreactive signal was obtained in this time group. After establiment in vitro decidualization of stromal cells, the homogenate from glan- dular epithelial cells was added into the decidual cell. The results revealed that the decidual cells became smaller when the homogenate was added for 3 days ,the PRL production in the 1μg/ml group was decreased but the PRL content in the control group was gradually augmented. These findings suggest that the decidualization of stromal cells can be inhibited by the glandular epithe- lial cells, and the pattern and mechanism need to be further studied.
