摘要
目的探讨LKB1基因在肝细胞性肝癌(HCC)中的表达及其与肝细胞癌患者预后的关系。方法采用免疫组化技术检测70例HCC患者肝癌及癌旁组织和20例正常肝组织中LKB1蛋白的表达,采用x2检验分析LKB1表达与临床病理因素的关系。对该组70例患者进行随访,并应用COX比例风险回归模型对其预后影响因素进行评估。结果 LKB1蛋白在HCC组织中的阳性表达率明显低于正常肝组织和癌旁组织(P<0.05)。HCC患者中,LKB1蛋白表达与HCC的分化程度,门静脉侵袭,TNM分期有关(P<0.05)。LKB1阳性表达的患者生存时间比阴性表达的显著延长(P<0.05)。门静脉侵袭、胆管癌栓、TNM分期和LKB1表达是影响HCC预后的独立危险因素(P<0.05)。结论 LKB1蛋白表达是影响HCC患者生存时间的独立因素之一,检测LKB1蛋白可能为HCC患者预后判断提供参考。
To explore the expression of LKB 1 in hepatocellular carcinoma (HCC), and investigate the relationship of expression of LKB 1 and prognosis of HCC patients. Methods LKB 1 expression was detected by immunohistochemistry in 70 HCC tissues, 70 adjacent-cancer tissues and 20 normal liver tissues. Correlation between the expression of LKB 1 and clinicopathological features was analyzed by x2 test. All of these 70 cases of HCC were followed-up and prognostic factors were evaluated by COX proportional hazards regression model. Results The rates of positive staining of LKB 1 protein in HCC tissues were significantly lower than those in the adjacent tissues and normal liver tissues (P〈0.05). The expression of LKB 1 in HCC was significantly correlated with histologic grade, vascular invasion and TNM stages (P〈0.05). Compared with the patients with negative expression of LKB 1 protein, patients with positive expression had significantly longer survival times. Vascular invasion, bile duct thrombi, TNM stages and LKB 1 expression level were independent risk factors of prognosis survival of HCC (P〈0.05). Conclusion LKB 1 expression is an independent factor of survival time in HCC. The expression of LKB 1 may be helpful for assessing the prognosis of HCC.
