摘要
目的:观察不同浓度氟化物对大鼠切牙成牙本质内质网伴侣分子表达的影响,探讨氟对成牙本质细胞的损伤机制。方法:随机分组的30只大鼠分别饮用氟化钠浓度为0、75、150×106的自来水,建立慢性氟中毒模型,采用SP免疫组化法检测大鼠成牙本质细胞中Grp78,XBP-1和Caspase-12的表达。利用MetaMorph显微图像分析软件采集数据并使用spss13.0软件分析。结果:随氟化物浓度增加,成牙本质细胞GRP78、XBP-1、Caspase-12的表达强度增加,且各组间具有显著差异(P<0.05)。结论:一定浓度的氟化物可以引起成牙本质细胞内质网应激,Grp78和XBP-1过表达,高强度的内质网应激会诱导成牙本质细胞通过活化Caspase-12导致成牙本质细胞凋亡。
Objective: To study the effect of different concentrations of fluoride on the expression of Endoplasmic reticulum chaperone, to explore the mechanism of odontoblast Damage on rats. Methods: 30 Wistar rats were divided into three groups randomly. Morphology and immunohistochemistry were used to detect the changes of the odontoblast and the expression of GRP78, XBP-1 and caspase-12 in rat incisors. Metamorph mieroscore images anal- ysis system and spss13.0 software were used to analyze the data. Results: The results of immunohistochemistry showed that GRP78, XBP-1, Caspase-12 were significantly different among the groups (P〈0.05). Conclusion: Excessive fluoride could induce odontoblast apoptosis and increase the expression of GRP78 and XBP-1, and sustained endoplasmic reticulum stress will lead to the odontoblast apoptosis through the activation of Caspase-12.
