miR-129-5p在胶质瘤中的表达及其对胶质瘤细胞糖代谢与生长的影响 被引量:7

Expression of miR-129-5p in glioma and its effect on glycometabolism and growth of human glioma cells

摘要 目的探讨微小RNA129-5p(miR-129-5p)在胶质瘤中的表达及其对人脑胶质瘤细胞糖代谢与生长的影响。方法采用RT-PCR技术检测miR-129-5p在胶质瘤组织中的表达水平。用miR-129-5p寡聚核苷酸转染胶质瘤细胞;通过MTT微量酶反应比色法、流式细胞术、葡萄糖和乳酸检测分析试剂盒,观察上调miR-129-5p表达后对胶质瘤细胞(U87)增殖、周期及糖代谢的影响。结果 miR-129-5p在胶质瘤组织中较正常脑组织呈低表达(P<0.01)。上调miR-129-5p的表达后,U87细胞的增殖受抑制,细胞周期阻滞在G1期,葡萄糖消耗量和乳酸生成量均降低(P<0.05)。结论 miR-129-5p在胶质瘤组织中呈低表达;上调其表达可以明显影响胶质瘤细胞的糖代谢和细胞的增殖能力。 Objective To study the expression of microRNA-129-5p(miR-129-5p) in glioma and its effect on the glycometaholism and growth of human glioma cells. Methods The expression of miR-129-Sp in human glioma was detected by RT-PCtL The miR-129-5p oligonucleotides were transfected into glioma cells (U87 cells). MTT assay, flow cytometry, glucose assay kit and lactate assay kit were used to evaluate the effect of miR-129-Sp expression on the proliferation, cell cycle and growth of U87 cells. Results The expression level of miR-129-Sp in glioma was lower than that in normal cerebral tissues(P〈0. 01). Upregulation of miR-129-Sp expression could suppress the growth of glioma cells, delay the cell cycle at G1 phrase, and decrease the glucose consumption and lactate production (P〈0. 05). Canclusion The expression of miR-129-Sp in human glioma is low. Upregulation of miR-129-Sp expression may significantly affect the glycometabolism and growth of human glioma cells.
