伊维菌素完全抗原的制备

摘要 为了制备伊维菌素完全抗原,试验对伊维菌素进行结构改造,使之成为带活性基团的半抗原,并进行质谱鉴定,然后分别采用N-羟基琥珀酰亚胺酯(NHS)法将半抗原与牛血清白蛋白(BSA)耦联,耦联物作为免疫抗原;采用混和酸酐法将半抗原与卵清白蛋白(OVA)耦联,合成包被抗原。结果表明:耦联成功,耦联物的结合比分别为31.6和22.6。说明试验成功制备了伊维菌素完全抗原。 To prepare the complete antigens of ivermectin. The structure of ivermectin was changed to become the haptens with actlvlty group, which were identified by mass spectrum. The haptens were conjugated to a bovine serum albumin (BSA) carrier protein as hnmunogen using N -hydroxysuccinimide ester method (NHS) , and conjugated to the ovalbumin OVA to synthesize the coating antigen using mixed acid an- hydride method. The results showed that the conjugation was successfully achieved, and the ratios of conjugation were 31.6 and 22.6, respec- tively. It indicates that the complete antigens of ivermetin are successfully prepared.
