橡胶树α-维管蛋白基因HbTUA1的克隆及表达分析 被引量:2

摘要 利用橡胶树胶乳EST文库克隆到一个TUA基因,命名为HbTUA1(GenBank登录号:KC333454)。该基因cDNA全长1 580 bp,其中5′UTR长45 bp,3′UTR长167 bp,编码区长1 368 bp,编码449个氨基酸。HbTUA1蛋白具有TUA类蛋白保守的GTP结合域。荧光定量PCR分析显示,HbTUA1基因在橡胶树胶乳中的表达量最高,其次是树皮和芽,而在种子和叶片中的表达量最低;在胶乳中,HbTUA1基因的表达显著受割胶和伤害诱导,在不同死皮程度的橡胶树中也存在明显差异。初步表明HbTUA1基因可能参与橡胶树胶乳再生与胁迫应答调控。 The α-tubulin (TUA) proteins are major components comprising microtubules,and play roles in multiple biological processes in plants.Previously,we have showed by proteomics techniques that a TUA protein was upregulated by tapping.Here,the full-length cDNA of a TUA gene was cloned for the first time from the latex of Hevea brasiliensis.The cDNA was named as HbTUA1,and submitted to GenBank under the accession no.KC333454.The 1 580 bp long cDNA consisted of a 45 bp 5'-UTR,a 167 bp 3'-UTR,and a 1 368 bp coding sequence that predicted a protein of 449 amino acids.The deduced HbTUA1 protein harbored a TUA-specific GTP-binding domain.The expression patterns of HbTUA1 were investigated by real-time quantitative PCR.Of the six tissues examined,HbTUA1 showed the highest expression in latex,then in the bark and bud,and the least in the seed and leaf.The expression of HbTUA1 in latex was induced by tapping and wounding,and differed among the rubber trees with different severities of tapping panel dryness.The results presented here suggested the involvement of HbTUA1 in the regulation of latex regeneration and stress responses in H.brasiliensis.
