摘要
有机氮源中氨基酸的种类及含量是影响抗生素发酵产量很重要的因素之一。对比了不同有机氮源对泰乐菌素发酵产量的影响,并利用Plackett-Burman实验设计对泰乐菌素发酵培养基的有机氮源进行优化。结果表明,鱼粉、玉米蛋白粉、甜菜碱是影响泰乐菌素产量的主要因素。当培养基中鱼粉含量为4.7g/L、玉米蛋白粉为10.5g/L、甜菜碱为0.06g/L时,泰乐菌素发酵产量较未优化前提高15%以上。同时对鱼粉和玉米蛋白粉中的氨基酸含量进行对比分析,发现泰乐菌素发酵培养基中适量添加鱼粉可使培养基中的氨基酸更加平衡,有利于泰乐菌素的合成。
The type and content of amino acids in organic nitrogen sources was a very important factor which affected the ti- ter of antibiotic fermentation. The effects of different organic nitrogen sources were compared,and organic nitrogen sources of tylosin fermentation culture were screened by Plackett-Burman design. The results showed that fish meal,corn gluten meal and betaine were the main factors affecting tylosin titer; when the fishmeal,corn gluten meal and betaine in medium reached 4.7, 10.5 and 0.06 g/L, respectively, the tylosin titer was improved by 15 %. According to the comparative analysis of the amino acid content of fish meal and corn gluten meal,adding some fish meal in the tylosin fermentation medium could make a more balance of amino acid and was beneficial to tylosin synthesis.
