摘要
为建立利用分光光度计测定猪精子密度的方法,本试验选择杜洛克、长白和大白种公猪各30头,利用分光光度计和血球计数板分别测定猪精液的吸光度和精子密度,建立二者的回归方程及利用分光光度计测定猪精子密度的技术,并对该技术进行验证。研究结果表明,精液吸光度和精子密度的最佳回归方程为y=3.3917X^(1.2499),分光光度计法和血球计数板法测定的猪精子密度显著相关(R=0.936),提示建立的分光光度计法可用于测定猪精子密度。
In order to establish the method for determining concentration of boar sperm with spectrophotometer, boars of Large White, Landrace and Duroc were used in this study. Absorbances of semen and sperm concentration were determined with spectrophotometer and hemacytometer, respectively, and then regression equation and technique for determining sperm concentration with spectrophotometer were es tablished, finally, the technique was verificated. The results showed that the best regression equation was y=3.3917X12499 and sperm concentration determined by spectrophotometer and hemacytometer was significantly relevant(R=0.936). These results indicated that the spectrophotometer technique established is feasible to use for determining sperm concentration of boar.
