摘要
目的探讨人参皂苷Re对糖尿病大鼠的心肌保护作用及机制。方法取40只Wistar大鼠高脂高糖饲料喂养,腹腔注射链脲佐菌素(STZ)35 mg/kg制作糖尿病模型,将34只造模成功大鼠随机分为模型组12只,人参皂苷11只,吡格列酮组11只,分别予以相应量纯净水、人参皂苷Re 25 mg/(kg·d)、吡格列酮10 mg/(kg·d)灌胃,连续给药4周;另取10只正常大鼠予标准饲料喂养4周作为对照组。干预4周后处死各组大鼠,分别检测血糖、血脂,血清和心肌组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)水平,并行心肌组织病理学观察。结果人参皂苷组和吡格列酮组空腹血糖和血清总胆固醇水平高于对照组,(P<0.01),低于模型组(P<0.01),两组间无统计学差异;人参皂苷组和吡格列酮组SOD活性低于对照组,MDA水平高于对照组(P均<0.05);血清和心肌组织中SOD活性均高于模型组(P<0.01)、MDA水平均低于模型组(P<0.05),但两组间无统计学差异。人参皂苷组和吡格列酮组心肌组织结构损伤明显轻于模型组,心肌细胞排列较模型组整齐。结论人参皂苷Re对糖尿病大鼠具有心肌保护作用;抗脂质过氧化作用是其作用机制之一。
Objective To investigate the myocardial protective effects of ginsenoside Re on rats with diabetes mellitus and the mechanism. Methods Forty healthy male Wistar rats were given streptozotocin (STZ) 35 mg/kg by intraperitone- al injection to make diabetic models. After 72 hours, 34 models were successful made. Then they were randomly divided into the diabetic model group (DM group, n = 12), ginsenoside Re group (Re group, n = 11 ) and pioglitazone group (P group, n = 11 ) and were given corresponding amount of pure water, ginsenoside Re 25 mg/( kg·d) and pioglitazone 10 mg/( kg·d) by intragastric administration for four weeks in a row. Another 10 healthy male Wistar rates were given stand- ard feed as the control group (control group). All rats were killed after 4 weeks. Glucose and lipids were tested. The su- peroxide dismutase (SOD) activities and contents of MDA in serum and myocardial tissues were detected respectively. Re- suits The levels of fasting blood glucose and serum total cholesterol in the Re and P groups were higher than those of the control group, lower than those of the DM group ( all P 〈 0.01 ), but no significant difference was found between the Re and P groups. Compared with the DM group, Re and P groups could increase the activity of SOD (P 〈 0.01 ) and decrease the content of MDA in serum and myocardial tissues (P 〈 0.05) and there was no significant difference between Re and P groups. Compared with the DM group, the myocardial injury was decreased, and myocardial cells arranged orderly in the Re and P groups. Conclusions Ginsenoside Re has myocardial protective effects on rats with diabetes mellitus. One of the mechanisms is the anti-lipid peroxidation.
