摘要
于 1 996年从长春地区一养鸡场分离到一株新城疫病毒 (NDV长春株 ) ,经过病毒生物学特性的研究表明该NDV毒株为弱毒株。将NDV长春株纯化培养后 ,提取病毒RNA ,经RT_PCR扩增F基因 ,并测定出F基因的全部核苷酸序列为1 75 8bp ,编码有 5 5 3个氨基酸残基。与国内外发表的部分NDV病毒的强毒株和弱毒株的相同序列进行比较 ,其核苷酸同源性在 88.2 %~ 96 .7%之间 ,氨基酸同源性在 90 .1 %~ 98.1 %之间 ,并与所有弱毒株F蛋白裂解位点区 (1 1 2~ 1 1 7)氨基酸序列Gly_Arg/Lys_Gln_Gly/Ser_Arg_Leu相同 。
The Changchun strain of Newcastle disease virus(NDV) was proved one avirulent by its biological characteristics.This study amplified the F gene of Changchun strain of NDV by RT_PCR and sequenced.The nucleotide sequening showed that the F gene of Changchun strain of NDV is 1758bp, encoding 553 anino acids residues and its homology of La Sota,D26,B1,Queensland,Ulster,Beaudette,Miyadera,Italian,F48,Australia_Victoria,Texas and Herts are 88.2%~96.7% and 90.1~98.1% separately.The amino acid sequences of cleavage site region(112~117)Gly_Arg_Gln_Gly_Arg_Leu are similar to that of all other avirulent strains.It was further confirmed that the Changchun strain of NDV is avirulent.
