亮菌多糖对顺铂致小鼠肠黏膜损伤的保护作用 被引量:8

Protective effect of polysaccharide from Armillariella tabescens on cisplatin-induced intestinal mucosal injury in mice

摘要 目的:探讨亮菌多糖(ATPS)对顺铂(DDP)致小鼠小肠黏膜损伤的保护作用。方法:90只雄性小鼠随机分为空白对照组,模型组,ATPS低、中、高剂量组(30,60,120 mg·kg-1),瑞巴派特(45 mg·kg-1)组;腹腔注射DDP 10 mg·kg-1建立小鼠小肠黏膜损伤模型,分别用HE染色和免疫组化观察小鼠小肠黏膜病理学改变与小肠黏膜增殖核抗原(PCNA)阳性细胞表达情况;采用ELISA法检测小鼠小肠黏膜组织表皮生长因子(EGF)含量。结果:与空白对照组比较,模型组小鼠小肠黏膜病理损伤积分显著增高;与模型组比较,ATPS 60,120 mg·kg-1组小肠病理损伤积分均显著降低。与模型组比较,ATPS 60,120 mg·kg-1组EGF含量和PCNA阳性细胞率也均显著增高;随着EGF含量增高,黏膜病理损伤和PCNA阳性细胞率分别有下降和增高趋势。EGF与PCNA阳性细胞率之间存在线性相关。结论:ATPS能刺激DDP致小鼠肠黏膜损伤后细胞增殖修复,对肠黏膜损伤起到保护作用,其机制与EGF调节有关。 Objective: To determine the protective effect of polysaccharide from Armillariella tabescens (ATPS) against cisplatin (DDP)-induced intestinal mucosal injttry in mice. Methods: Male mice (n = 90) was randomly divided into groups, and treated with ATPS (30, 60 and 120 rag kg-l) or rebamipide (45 rag kg-l). Intestinal mucosal injury was induced by single intraperitoneal injection of DDP 10 mg. kg-1. The pathological changes and expression of proliferative cell nuclear antigen (PCNA) were determined by HE staining and immuno- histochemistry, respectively, in small intestinal mucosa. The levels of EGF in small intestinal samples were assayed by ELISA. Results: The pathological scores in small intestinal samples were significantly increased in injured mice compared with control mice. ATPS at 60 and 120 mg" kg-I significantly reduced the pathological scores, and signif- icantly increased the levels of EGF and the expression of PCNA. The pathological scores decreased and the expres- sion of PCNA increased with the increase in the levels of EGF in small intestinal samples. The expression of PCNA had linear correlation with levels of EGF. Conclusion: ATPS attenuates cisplatin-induced intestinal mueosal injury in mice by stimulating mucosal cell proliferation via mechanisms involving EGF.
