2种缓冲液对血清蛋白醋酸纤维膜电泳效果对比研究 被引量:3

摘要 目的在血清蛋白醋酸纤维膜电泳巴比妥缓冲液可以分离出5条区带的实验条件下,研究硼酸-硼酸盐缓冲液是否能够达到相同甚至更好的实验效果,是否可替代巴比妥缓冲液。方法采用DYY-8C型电泳仪进行醋酸纤维膜电泳,选择不同电压和电泳时间,观察巴比妥缓冲液与硼酸-硼酸盐缓冲液分离兔血清蛋白的效果,并分析2组蛋白质相对含量测定值有无差异。结果在相同的实验条件下,硼酸盐缓冲液的分离效果好于巴比妥缓冲液,2组蛋白质相对含量无明显差异。结论在血清蛋白醋酸纤维膜电泳实验中,与巴比妥缓冲液相比,硼酸缓冲液的实验结果更稳定且重复性更好,可以替代巴比妥缓冲液。 Objective Boric acid - borate buffer can achieve the same or even better experimental effect in the experimental conditions that barbiturate buffer can separate out five ribbons in serum protein cellulose acetate membrane electrophoresis. Methods Using DYY-SC electrophoresis apparatus for cellulose acetate membrane electrophoresis, choosing different voltage and time, the effect separating rabbit serum protein is observed in barbiturate buffer and boric acid - borate buffer. And the relative content of protein components is measured. Results Under the same experimental conditions, the effect separating serum protein in borate saline buffer is better than in barbiturate buffer. There is no difference between the two kinds of buffer. Concluaiona In serum protein cellulose acetate membrane electrophoresis, the boric acid - borate buffer can replace barbiturate buffer, and many experiments prove that : the experimental results of boric acid - borate buffer is more stable and repeatability is better than barbiturate buffer.
