黄芪对胆道梗阻后心肌损伤保护作用的实验研究

Experimental Study on Myocardial Injury and Protective Effect of A stragalus Membranaceus after Biliary Obstruction

目的 :观察胆道梗阻后心肌损伤及黄芪 (AM)的保护作用。 方法 :动态观测心肌组织丙二醛 (MDA)和超氧化物歧化酶 (SOD)、血清T Bil、TBA、内毒素 (ET)、肿瘤坏死因子 (TNFα)含量 ;取左室心肌行光电镜检查 ,应用ABC免疫组化染色法 ,定位TNFα 在心肌组织中的表达和分布。 结果 :胆道梗阻后 ,血清T Bil、TBA、ET、TNFα 水平逐渐升高 ,心肌组织MDA含量逐渐升高 ,SOD逐渐减少 ;各AM治疗组与同时相梗阻组比较 ,血清ET、TNFα、心肌组织MDA含量减少 ,SOD含量升高 ;心肌组织MDA含量与血清TNFα 含量变化呈正相关 ;光电镜下可见随胆道梗阻时间延长 ,心肌组织细胞损害加重 ,各AM治疗组与同时相梗阻组比较组织细胞损害减轻。 结论 :AM通过对抗自由基损伤、内毒素血症、TNFα 的综合作用 。

To investigate myocardial injury and the p rotective effects of astragalus membranaceus(AM) after biliary obstruction [W T5HZ]Methods:The contents of myocardial malondialdyhyde(MDA) and superoxide dismutase(SOD),the levels of serum T-Bil,TBA,endotoxin(ET), tumor necrosis factor(TNF α) were determined The left ventricle were drawn fir lig ht and electronic microscopic observation. Immunohisto-chemical staining method of ABC was to locate the expression and distribution of TNF α in myocardial ti ssues.Results:After ligation of the common bile duct serum T Bil, TBA, ET, TNF α levels and myocardium MDA were increased signifi cantly,while SOD decreased markedly as compared with the control group. As compa red with BDL group at the same phase ,serum T Bil, TBA, ET, and TNF α levels and myocardium MDA in AM-treated groups were decreased significantly,while myoca rdium SOD increased. The myocardium MDA content was highly positively correlated with serum TNF α levels. There were less myocrdial injuries in AM treated gr oups than that in BDL group at the same stage with ilght and electronic microsco pic observation.Conclusion:AM can protect myocardium effectively from the injury after biliary obstruction against ET,TNF α and my ocardial free radical injury.
