葛根素抗ApoE^(-/-)小鼠动脉粥样硬化的炎症机制研究 被引量:11

The inflammatory mechanism of puerarin anti-atherosclerosis in ApoE knockout mice

摘要 目的:观察葛根素对ApoE基因敲除小鼠炎症因子的影响,探讨其是否能抗AS炎症反应;方法:将24只ApoE-/-小鼠随机分为模型组、葛根素组、辛伐他汀组,每组8只,另取8只C57BL/6J小鼠作为正常对照组。采用高脂喂养16周的方式建立AS模型,分别以胸主动脉与主动脉窦部位切片观察主动脉斑块形态,荧光定量PCR法检测小鼠主动脉斑块内IL-2、IL-6、TNF-α、hsCRP等炎症因子mRNA表达;结果:与正常对照组相比,模型组小鼠血脂与炎症因子mRNA水平明显增高,主动脉内有纤维斑块形成;辛伐他汀(5mg/kg)与葛根素(65mg/kg)均能明显降低炎症因子mRNA表达,并且能抑制斑块形成,葛根素效果更佳。结论:葛根素能降低AS斑块炎症因子的表达,抗炎机制可能是其抗AS的其中一个药理机制。 To study whether puerarin can anti-inflammatory response in atheroscleresis; Methods :24 ApoE^-/- mice were randomly divided into model group, puerarin group, simvastatin group (n =8), another 8 C57BL/6J mice served as normal group. High fat diet was giving to ApoE/ mice for 16 weeks to establish AS model. The thoracic aorta and aortic sinus was sliced to observe aortic plaque morphology; Quanti tative PCR assay was used to detect IL-2, IL-6, TNF-^1, hs-CRP mRNA in mouse aortic; Results:compared with the normal control group, the mRNA levels of inflammatory cytokines and blood lipids in model mice was significantly higher ( P 〈 0.01 ), and there was intra-aortic fi brous plaque formation; Sim atorvastatin and puerarin could significantly not only reduce the inflammatory cytokine mRNA expression ( P 〈 0. 01 ), but also inhibit plaque formation, especially in puerarin group; Condusion:Puerarin can reduce the expression of inflammatory factors in AS plaque, which may be one of pharmacological mechanisms of puerarin anti-atherosclerosis.
