瘤果黑种草子挥发油对慢性阻塞性肺病大鼠肺部炎症的影响 被引量:5

Effect of volatile oil from Nigella glandulifera on lung inflammation of chronic obstructive pulmonary disease rats model

摘要 目的:观察瘤果黑种草子挥发油对慢性阻塞性肺病(COPD)大鼠模型肺部炎症的影响。方法:48只SD大鼠,随机分为6组:正常对照组、模型组、氨茶碱组、瘤果黑种草子水煎液组、瘤果黑种草子挥发油低剂量和高剂量组。采用气管内滴入脂多糖(LPS)后雾化吸入木瓜蛋白酶建立COPD大鼠模型,28d后分别灌胃给药治疗,正常组和模型组给予生理盐水,每天给药1次,连续给药20d。观察各组大鼠的一般状况,HE染色观察肺组织的病理形态学改变,取支气管肺泡灌洗液(BALF)进行炎性细胞分类,酶联免疫法测定BALF上清液IL-8、TNF-α和NF-κB的含量水平。结果:瘤果黑种草子挥发油0.07、0.28 ml/kg剂量可明显改善COPD模型大鼠的一般状况,对其肺组织的病理变化有不同程度的减轻;BALF中的巨噬细胞、中性粒细胞所占比例显著下降,IL-8、TNF-α和NF-κB的含量水平均降低。结论:瘤果黑种草子挥发油有一定的抗COPD大鼠模型肺部炎症作用,其机制可能与减弱炎性细胞对LPS的反应,抑制炎症因子的合成和释放有关。 To investigate the effect of volatile oil from Nigeria gland ulifera on lung inflammation of chronic obstructive pulmonary dis ease (COPD) rats model. Method: Forty-eight SD male rats were randomly divided into: normal group, model group, aminophylline treat ment group, water decoction treatment group, VONG low dose treatment group, and VONG high dose treatment group. The COPD rat model was established by intratracheal Instillation of lipopolysaccharides (LPS) with inhalation of papain. After 28 d, each group rats with treatment by ig administration, once daily for twenty days, and rats in the normal group and model group with same volume of normal saline. The gener al situation of rats were observed and the rats lung tissue were obtained and stained with HE for histopathological examination, inflammatory cells in bronchuoalveolus lavage fluid (BALF) were classificatied, the levels of inflam- matory factors, produced by inflammatory cells, of IL-8 ,TNF-α and NF-KB in BALF were measured by ELISA. Result:VONG 0.07,0.28 ml/kg could improve the general condition of COPD rats, and the pathologic changes of lung tissue were reduced in different degree; the macrophage cells, neuttmphil cells percentage in BALF were decreased ( P 〈 0.01 ), and levels of IL-8 ,TNF-α and NF-KB in BALF were decreased ( P 〈 0.01 ) significantly. Conclusion: VONG shows some anti-inflammatory effect in COPD model rat lung, The mechanisms might be related to the reduction of reaction of inflammatory cells to LPS, and inhibition of synthesis and release of inflammatory cytokines.
