复方栀子凝胶对银屑病动物模型的干预作用 被引量:5

Effect of fufangzhizi gelatin on mouse psoriasis-like lesions

摘要 目的:本实验旨在从普萘洛尔和咪喹莫特诱导的银屑病样模型中优选出一种银屑病模型,并考察复方栀子凝胶的影响。方法:采用药物诱导的方法激发小鼠皮肤出现银屑病样的变化,并通过小鼠皮肤病损的(PASI)评分来观察银屑病样小鼠模型皮损的变化情况,光镜下观察皮损组织形态学变化,测量表皮厚度。免疫组织化学法检测皮损皮肤中增殖细胞核抗原(PCNA)。综合各指标结果,优选出一种模型作为药物疗效考察的银屑病动物模型,在此模型和鼠尾鳞片颗粒模型的基础上考察复方栀子凝胶对银屑病的影响。复方栀子凝胶确定起始剂量为0.1g/g,考察剂量范围为0.1g^0.2g/g。结果:咪喹莫特诱导的动物模型小鼠背部皮肤出现红斑、表皮肥厚、鳞屑成层等现象;而普萘洛尔诱导的动物模型无此种状况出现,另外普萘洛尔与咪喹莫特诱导的小鼠模型均有角化不全、表皮棘层增厚的现象。使用高(0.2g/g)、中(0.15g/g)、低(0.1g/g)剂量复方栀子凝胶之后,咪喹莫特银屑病模型的症状得到明显改善,银屑病样皮损减轻,PASI评分降低,角化不全现象得到改善,表皮厚度明显薄于模型对照组和阴性对照组,其中高剂量组改善更加明显。复方栀子凝胶0.2g/g组、0.15g/g组、0.1g/g组与正常组相比较对小鼠鳞片表皮颗粒层均有明显的促进作用,高剂量组更加明显。结论:咪喹莫特诱导的银屑病样模型更接近临床症状,复方栀子凝胶对银屑病有较好的影响。 This study is to select a a psoriasis model from two mouse models by drug-induced and to observe the effects of fufangzhizi gelatin on mouse psoriasis -like lesions induced by imiquimod. Methods: This method is to emerge psoriatic skin changes in skin of mouse by drug - induced. The lesions were evaluated according to the psoriasis area and severity index(PASI). The histology and epidermal thiekeness were observed under light microscope. The expression of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemieal staining. Select a better psoriasis model by the result of comprehensive index as a mouse psoriasis . Observe the effects of fufangzhizi gelatin by mouse model induced by imiquimod and number of scale with granular layer in tail epidermis scale in mice . Effective dose of fufangzzhizi gel is 0. 15g/g. We determine the origination dose is 0. lg/g and the dose range of 0. lg -0.2g/g. Results:The results show that models induced by imiquimod is more similar to real psoriasis-like lesions with the symptoms of erythema and epidermal thickening ,while the models induced by propranolol without the symptom. Both of the models induced by imiquimod and propranolol have the symptoms of parakeratesis and thicken ing epidermal stratum spinosum. Compared with the model, the cutaneous symptoms were alleviated, PASI scores decreased , epidermal para keratosis relived ,epidermal thickness obviously thinner than the compared groups after applying fufangzhizi gelatin. It showed that the forma tion of the granular layers in the flakes of the mouse-tails was facilitated in Fufangzhizi gelatin groups. Conclusion: This experiment has select ed a psoriasis -like lesions model induced by imiqulmod and proved that fufangzhizi gelatin do have therapeutic effect on psoriasis like lesions as well as alleviate the symptoms.
