摘要
目的探讨上皮钙黏附蛋白(E-cadherin)、神经型钙黏附蛋白(N-cadherin)、α-连环蛋白(α-catenin)、β-连环蛋白(β-catenin)在非小细胞肺癌(NSCLC)中表达的意义。方法采用免疫组化方法结合计算机图像分析技术,检测E-cadherin、N-cadherin、α-catenin、β-catenin及Ki-67在139例NSCLC组织、43例癌旁组织和39例正常肺组织中的表达水平和β-catenin在细胞中的分布变化。结果①NSCLC与正常肺组织相比,E-cadherin和α-catenin表达量显著降低(P<0.01),β-catenin表达量显著升高(P<0.01);NSCLC与癌旁肺组织相比,E-cadherin和α-catenin表达量明显降低(P<0.05);癌旁肺组织与正常肺组织相比,β-catenin表达量明显升高(P<0.05)。②E-cadherin、α-catenin的表达与NSCLC的分化程度相关,随着分化程度的降低,表达减弱(P<0.05);低分化与高、中分化NSCLC相比,N-cadherin(P<0.01)、β-catenin(P<0.05)表达量明显升高。③随着NSCLC分化程度的降低,β-catenin核表达水平显著升高(P<0.01);在有淋巴结转移(P<0.05)和具有较高增殖能力的NSCLC组织(P<0.01)中,β-catenin核表达水平显著升高。结论①E-cadherin、α-catenin表达下调与NSCLC细胞分化程度直接相关。②钙黏附蛋白表型转化与NSCLC细胞的分化程度密切相关。③β-catenin降解抑制与NSCLC的发生有密切关系。④β-catenin核表达与NSCLC的分化、增殖、转移具有密切关系。
Objective To study the significance of E-cadherin, N-cadhefin, α-catenin, β-catenin and Ki-67 expression in non-small cell lung cancer (NSCLC). Methods A total of 139 NSCLC cases, 43 tumor-adjacent tissues and 39 normal lung tissues were involved in this study. The expression of E-cadherin, N-cadherin, α-catenin, β-catenin, Ki67 and subcellular location of β-catenin were detected by using immunohistochemistry and computer-imaglng analysis. Results There was a significant decrease in the expression of E-cadherin (P 〈 0. 01 ), α-catenin (P 〈 0. 01 ) and significant increase in β-catenin (P 〈 0. 01 ) in NSCLC compared with normal lung tissues. Decreased E-cadherin ( P 〈 0. 05 ) and α-catenin expression (P 〈 0. 05)were detected in NSCLC as compared with tumor-adjacent tissues. Only the expression of β-catenin (P 〈 0. 05) was significantly increased in tumor-adjacent tissues as compared with normal lung tissues. The expression of E- cadherin and α-catenin was significantly associated with NSCLC differentiation ( P 〈 0. 05 ) ; Increased N-cadherin (P 〈 0. 05 ) and β-catenin (P 〈 0. 05 ) expression was detected in poorly differentiated NSCLC as compared with moderately and well differentiated NSCLC; Increased β-catenin nuclear expression was detected from well, moderately to poorly differentiated NSCLC (P 〈 0. 01 ) ; In NSCLC with lymph node metastasis (P 〈 0. 05) and higher reproductive activity (P 〈 0. 01 ), β-catenin nuclear expression was also significantly increased. Condusions The down-regulation of E-Cadherin and α- Catenin is significantly associated with the degree of NSCLC differentiation, and so does cadherin switch. Inhibition of β-catenin degradation is significantly associated with the carcinogenesis of NSCLC. In addition, β-catenin nuclear expression is also significantly associated with the differentiation, proliferation, and metastasis of NSCLC.
