油桐叶片愈伤组织诱导及植株再生 被引量:14

Callus Induction from Leaves and Plant Regeneration of Tung Tree(Vernicia fordii Hemsley)

摘要 以油桐无菌苗叶片为试材,研究不同种类及浓度的植物生长调节剂对愈伤组织诱导、分化、增殖及生根的影响。结果表明:叶片愈伤组织的最佳诱导培养基为1/2MS+2.0 mg·L-16-BA+1.0 mg·L-12,4-D,诱导率达100%;最佳分化培养基为1/2MS+3.0 mg·L-16-BA+0.1 mg·L-1IBA+0.05 mg·L-1IAA,分化率为86.36%;最佳继代增殖培养基为MS+3.0 mg·L-16-BA+0.05mg·L-1IBA;最佳生根培养基为1/2MS+0.1 mg·L-1IBA,生根率93.83%。炼苗后移栽到泥炭土:珍珠岩:蛭石=2:1:1的基质中,成活率达92%以上。 The effects of different concentrations and combinations of plant growth regulators on callus induction, shoot differentiation, proliferation and root formation were studied using germ-free leaves of tung tree (Vernicia fordii) as plant materials. The results showed that the fittest medium for callus induction from leaves were 1/2MS+2.0 mg.L-1 6-BA+I.0 mg-L-1 2,4-D, with a 100% callus induction rate; the fittest medium for shoot differentiation were 1/2MS+3.0 mg.L-1 6-BA+0.1 mg.L-1 IBA+0.05 mg.L-1 IAA, with a 86.36% differentiation rate; the best medium for shoot multiplication was MS+3.0 mg'L-16-BA+0.05 mg'L1 IBA; the best rooting medium was 1/2MS+0.1 mg-L-1 IBA, with a 93.83% rooting rate. After the culture-bottle seedlings were transplanted into the matrix composed of peat soil, perlite and vermiculite (2:1:1), the survival rate was up to 92%.
