摘要
研究赤霞珠葡萄叶不同溶剂萃取物中黄酮和总酚的含量及抗氧化活性。用不同极性溶剂依次对赤霞珠葡萄叶φ=95%乙醇粗提物进行萃取,分别得到石油醚萃取物、氯仿萃取物、乙酸乙酯萃取物、正丁醇萃取物、水萃取物及经大孔树脂φ=70%乙醇洗脱物。测定不同萃取物中黄酮和总酚的含量,研究不同萃取物对DPPH自由基、ABTS自由基、超氧阴离子自由基(O2-·)的清除能力及还原力,并与BHT进行比较,评价其抗氧化活性。赤霞珠葡萄叶不同溶剂萃取物均具有一定的抗氧化活性,且随着浓度的增加而增强。φ=70%乙醇洗脱物和乙酸乙酯萃取物均具有较强的抗氧化活性,其清除DPPH自由基和O2-·的IC50分别为(0.017±0.001)、(0.023±0.001)g/L和(0.174±0.007)、(0.222±0.003)g/L,高于BHT。相关分析表明,不同溶剂萃取物中黄酮、总酚含量与抗氧化能力具有相关性。赤霞珠葡萄叶萃取物具有很好的抗氧化活性,不同溶剂萃取物对抗氧化活性有很大影响。
In order to study the antioxidant activities of the total phenolics and flavonoids contents of different solvent extracts from Cabernet sauvignon leaves, 95% ethanol crude extract from Cabernet sauvignon leaves were successively extracted by using different solvents to provide petroleum ether extract, chloroform extract, ethyl acetate extract, n butyl alcohol extract, water extract and 70% ethanol elution. The contents of total phenolics and flavonoids in different extracts were detected. Different extracts on DPPH radical, ABTS cation radical, superoxide anion free radical scavenging ability and reducing power were analyzed, and it was compared with butylated hydroxytoluene to evaluate its antioxidant activity. Cabernet sauvignon leaves extracts from different solvents had certain antioxidant activity. The activity increased with the increase of the concentration. 70% ethanol elution sample and ethyl acetate extract showed remarkable scavenging activity on DPPH IC50 (0.017±0.001), (0.023±0.001) g/L, O2-[KG-*3/4]· IC50 (0.174±0.007), (0.222±0.003) g/L, higher than that of butylated hydroxytoluene. Correlation analysis showed that extracted total flavonoids, phenolic content in different solvents had correlations with antioxidant capacity. Cabernet sauvignon leaves extracts had stronger antioxidant activity, and different polar solvents had greater influence on antioxidant activity.
