摘要
以非酶糖基化反应抑制率为评价指标,采用单因素实验筛选影响猴头菌(Hericium erinaceus)发酵液活性的培养基成分,结果表明,①木糖替换PD培养基中葡萄糖,②红豆或绿豆替换PD中马铃薯,③PD添加酵母浸出汁、NaH2PO4·2H2O、维生素B2均可提高抑制率;并在此基础上利用响应面和BoxBehnken Design进一步优化猴头菌液态发酵培养基,当培养基组成为240.50g/L红豆,173.41g/L绿豆,19.49g/L木糖,4.72g/L酵母浸出汁,1.30g/L NaH2PO4·2H2O,56.89mg/L维生素B2,用于猴头920发酵,发酵液对非酶糖基化反应抑制率为(88.51±3.23)%,基本与模型预测值相符,较未优化提高了214%。
Selected components of Hericium erinaceus culture medium affecting the ability of the spent culture fluid to inhibit non-enzymatic glycosylation (NEG) were optimized using response surface methodology based on single factor tests. In single factor tests, xylose, yeast extract, NaH2PO4 * 2HzO, vitamin B2, Red beans and Mung beans were evaluated as potential key factors affecting NEG inhibition. Maximum inhibition (88.51%) was recorded when a growth medium consisting of (g/L) : 240.5 red beans, 173.41 Mung beans, 19.49 xylose, 4.72 yeast, 1.30 NaHz PO4 2H2O and 0. 05689 vitamin B2was used. This value was 214% higher compared with potato dextrose (200 g/L potato, 20 g/L glucose) controls.
