缺氧对小鼠脂肪细胞分泌的细胞因子的影响 被引量:1

摘要 目的探讨缺氧状态下,成熟小鼠前脂肪细胞(3T3-L1)分泌的肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、脂联素的变化。方法将3T3-L1细胞诱导为成熟的脂肪细胞后,随机分为正常组和缺氧组,每组分为4 h,8 h,16 h,24 h共4个时间点,每个时间点细胞细胞孔数为6。用酶联免疫分析法(ELISA)测定细胞培养液中4 h、8 h、16h、24 h各时间点缺氧处理前后TNF-α、IL-6、脂联素的含量变化。结果与相应时间点正常组相比,缺氧使IL-6含量明显上升(P<0.05),脂联素含量下降(P<0.05),而TNF-α表达水平变化不大(P>0.05)。结论缺氧抑制脂肪细胞分泌抗炎因子脂联素的表达,上调炎症因子IL-6的表达,提示缺氧可能在炎症的发生过程中起着重要的作用。 Objective In order to study the influence of hypoxia on secretion of interleukin - 6 ( IL - 6 ), TNF-α and adiponectin in 3T3 - L1 adipocytes from adult mice. Methods 3T3 - L1 adipocytes were induced into mature adipocytes after treatment with hypoxia for 4 h, 8 h, 16 h and 24 h.. The culture medium was collected and tested for interleukin - 6 ( IL - 6), TNF -α and adiponectin by using enzyme - linked immunosorbent assay (ELISA). Results In comparison with normal condition, the concentration of IL - 6 was obviously increased ( P 〈 0.05 ), the concentration of adiponectin was reduced ( P 〈 0.05 ) and level of TNF -α was not altered ( P 〉 0.05 ) by hypoxia. Conclusion Hypoxia can down - regulate the secretion of anti - inflammatory adipokine - adiponectin, and up - regulated the secretion of inflammatory adipokine - IL - 6, thus it suggests that hypoxia is correlated with inflammatory response.
