摘要
研究辣木种子提取液中蛋白质组成,采用净水活性追踪法,确定辣木种子中具有絮凝活性成分的蛋白质。结果表明:辣木水提液和盐提液中都是由等电点为2.42、3.41和11.38的3种主要蛋白质组成。在水提液中,等电点为3.41的蛋白质含量最高、达75.7%,其余2种含量分别为4.2%、20.1%;在盐提液中,等电点为11.58的蛋白质含量最高、达65.2%,其余两种含量分别为20.8%、14.0%。净水活性试验结果表明,辣木提取液中等电点为2.42、3.41的2种蛋白质净水活性差,不是辣木絮凝的活性成分;等电点为11.38的蛋白质具有较高的净水活性,絮凝5 h后的净水活性达93.4%,是辣木絮凝活性最主要的蛋白质成分。
Protein composition of Moringa ole^era seed extract was studied, and proteins with flocculent active ingredients were determined in M. oleifera seeds by bioassay-guided method. The results showed that the water extract and salt extract were made up of three major proteins whose isoelectric point were 2.42, 3.41 and 11.38, respectively. The content of protein with isoelectric point 3.41 was 75.7%, the contents of other two proteins were 4.2% and 20.1% respectively in the water extract; The content of protein with isoelectric point 11.58 was 65.2%, the contents of other two proteins were 20.8% and 14.0% respectively in the salt extract. The activity tests showed that, the proteins with isoelectric points 2.42 and 3.41 respectively had poor activities, so they were not the active ingredients of Moringa flocculation; The protein with isoelectric point 11.38 had high flocculating activity, its flocculating activity was up to 93.4% after 5 hour floeculaion, therefore this protein was the main component of flocculating activity of M. oleifera.
