摘要
采用石墨炉原子吸收光谱法进行细胞中微量金属元素铁、铜、锰含量的测定。分别针对贴壁培养的成骨细胞MC3T3-E1和悬浮培养的白血病细胞K562进行量化分析,依据细胞总数和细胞内总蛋白含量进行归一化处理,所得结果与文献报道具有较好的一致性。该方法灵敏度高,重复性好,适用于培养细胞中铁、铜、锰等微量金属元素的定量测定,为微量金属元素在细胞中的作用以及细胞组分研究提供实验依据。
In this paper, the contents of trace metal elements iron, copper and manganese in cells were detec- ted by graphite furnace atomic absorption spectrometry. For different cells including adhere-wall cultured osteo- blast MC3T3:E1 and Suspension cultured leukemia cell K562, the quantitative analysis was conducted by the normalization based on total number and total protein content respectively. The results showed good consisten- cy with those of literature. In summary, this method was suitable for determination of the contents of Fe, Cu and Mn in the cultured cells with high sensitivity and good repeatability,and provided experimental basis for the research of the role of trace metal elements in cells as well as the cell components.
