摘要
目的建立高效液相色谱-质谱联用测定人血浆中麦考酚酸浓度的方法。方法以苯巴比妥为内标,色谱柱为ZORBAX Eclipse XDB—C18(2.1mm×150mm,3.5μm),流动相为0.05%乙酸铵-乙腈(45:55),流速为0.2mL·min^-1,柱温35℃,电喷雾离子源(ESI)正负离子同时检测。结果麦考酚酸的线性范围为0.5—100.0μg·mL^-1;最低检测限分别为0.1μg·mL^-1。高、中、低3个浓度的日内标准偏差小于3%,日间标准偏差小于15%,绝对回收率均大于70%。结论本方法简便、快速、灵敏、重现性好,能有效检测人血浆中麦考酚酸的含量。
Objective To establish a HPLC -MS method for determina- tion the concentration of mycophenolic acid in human plasma. Methods Phenobarbital was used as internal standard. After precipitation, the analysis was chromatographed on a XDB - C18 (2. 1 mm ×150mm,3.5μm). An isocratic mobile phase consisted of 0. 05% ammonium acetate in water - aceto- nitrile (45:55). The flow rate was 0. 2 mL· min^-1 and column temperature was 35 ℃. ESI electrospmy ionization source in both negative and positive modes. Results The calibration curves of each drug showed a good linearity in a range of concentration between 0. 5 - 100. 0 μg·mL^-1 for sodium valproate. Intra and inter precision(RSD% ) were less than 3% and 15% , respectively. The recoveries were above 70%. Conclusion The method is simple, rapid, sensitive and stable which is suitable for determination of the concentration of mycophenolic acid in human plasma.
