摘要 目的观察白术多糖(PAM)对小鼠Kupffer细胞免疫功能的影响。方法 PAM体外作用于正常Kupffer细胞,以吞噬中性红A540、分泌NO和TNF-α的量以及LDH漏出量评价其免疫功能及细胞受损害情况。小鼠ig PAM后,检测Kupffer细胞的上述指标及胞内酸性磷酸酶(ACP)活性及血清ALT和GST。结果 PAM增强Kupffer细胞对中性红吞噬作用、增加ACP活性及NO和TNF-α的产生。PAM在体外不增加肝实质细胞LDH漏出,对sALT和sGST的活性没有影响。结论 PAM可激活Kupffer细胞免疫功能,增强机体抵抗能力。 Objective To observe the influence of polysaccharide of A tractylodes macrocephala Koidz (PAM ) on the immunologic function of mice Kupffer cells .Methods Normal mice Kupffer cells were treated with PAM in vitro .Absorbance at 540 nm (A540 ) of neutral red absorption and supernatant NO ,TNF-αin the cells were measured to evaluate the immunologic function of Kupffer cells ;LDH leakage was measured to estimate the severity of cellular damage .The above indices and ACP of Kupffer cells and sGST and sALT activity were measured after mice were given PAM in vivo .Results PAM markedly enhanced the phagocytic activity ,ACP and supernatant NO ,TNF-αof Kupffer cells both in vitro and in vivo .The LDH leakage of the hepatocytes and the sGST and sALT activity were not increased in vitro .Conclusion PAM with suitable dose could activate the function of Kupffer cells .PAM could improve the body's resistance to disease and has a practical application in the treatment of immunodeficiency dis-ease ,cancer and other diseases .
