扁桃茎段增殖培养基的筛选

摘要 以"莎车14号"、"莎车大3号"2个扁桃品种的茎段为试材,采用L9(34)正交实验设计,研究了适合"莎车14号"、"莎车大3号"茎段增殖的培养基。结果表明:外植体灭菌以70%乙醇消毒8s+2%和4%次氯酸钠消毒10min效果良好,茎段平均成活率为95%。扁桃"莎车大3号"的最优增殖培养基配比为MS+6-BA 1.0mg/L+IAA 0.1mg/L,增殖系数4.63;扁桃"莎车14号"的最优增殖培养基配比为MS+6-BA 1.0mg/L+IAA 0.2mg/L+GA30.1mg/L,增殖系数4.20。 Selecting almond cultivars ' Shacheda3 ', ' Shache14 ~ stem as materials, the ideal multiplying medium of them were screened through the orthogonal test of L9 (34). The results showed that stems disinfection effect treated by 2%- 4 %NaClO for 10 rain,after 70% ethanol for 8 seconds was the best,and the average survival rate could reach 95%. The optimum medium of multiplication for ' Shacheda3' was MS-% 6-BA 1.0 rag/L+ IAA 0. 1 mg/L, and the multiplication rate was 4. 63;the optimum medium of multiplication for 'Shache 14' was MS+6-BA 1.0 rng/L+IAA 0. 2 mg/L-I-GA3 0. 1 mg/L,and the multiplication rate was 4. 20.
