摘要
目的观察电针对去卵巢大鼠模型肠黏膜钙结合蛋白-D9k(CaBP-D9K)mRNA和蛋白表达的影响,探讨针灸治疗绝经后骨质疏松症可能机制。方法将60只3月龄SD健康雌性大鼠随机分为手术组和假手术组,3个月造模成功后将手术组随机分为模型组、假电针组、电针组、药物组。干预治疗12星期后,测定腰椎及股骨骨密度(BMD),然后处死大鼠,提取小肠黏膜,采用荧光实时定量PCR和Western blotting检测CaBP-D9K mRNA和蛋白表达。结果成功复制绝经后骨质疏松症模型,经电针组和药物组干预治疗12星期后,其大鼠骨密度与模型组相比均有明显提高,荧光实时定量PCR和Western blotting检测结果显示电针组和药物组大鼠小肠黏膜CaBP-D9K mRNA和蛋白表达上调,但电针组上调CaBP-D9K mRNA的水平不及药物组。结论针灸治疗绝经后骨质疏松症机制之一可能与上调小肠黏膜CaBP-D9K mRNA和蛋白表达量,从而增强肠钙吸收有关。
Objective To investigate the effect of electroacupuncture on the expressions of calcium binding protein-D9k (CaBP-D9K) mRNA and protein in intestinal mucosa of ovariectomized rats and explore a possible mechanism for acupuncture treatment of postmenopausal osteoporosis. Methods Sixty 3-month-old healthy female SD rats were randomly allocated to operation and sham operation groups. Following successful model making after 3 months, the operation group was randomly allocated to model, sham electroacupuncture, electroacupuncture and medication groups. Lumbar vertebral and femoral bone mineral density (BMD) was measured after 12 weeks of interventional treatment. The rats were then sacrificed. Small intestinal mucosa was taken. The expressions of CaBP-D9K mRNA and protein were examined using real time fluorescence quantitative PCR and Western blotting. Results A model ofpostmenopausal osteoporosis was made successfully. After 12 weeks ofinterventional treatment, bone mineral density was inc,eased significantly in the electroacupuncture and medication groups compared with the model group. The results of measurement using real time fluorescence quantitative PCR and Western blotting showed that the expressions of CaBP-D9K mRNA and protein in small intestinal mucosa were up-regulated in the electroacupuncture and medication groups of rats, but the level of up-regulation of CaBP-D9K mRNA was lower in the electroacupuncture group than in the medication group. Conclusion One of the mechanisms of acupuncture treatment for postmenopausal osteoporosis may be that up-regulated expressions ofCaBP-D9K mRNA and protein in small intestinal mucosa increase intestinal absorption of calcium.
