星形细胞肿瘤发生的分子生物学机制及与DEK mRNA表达的相关性研究

摘要 目的探讨DEK mRNA的表达与星形细胞肿瘤发生、发展的关系及星形细胞肿瘤发生的分子生物学机制。方法应用RT PCR技术检测各级别星形细胞肿瘤组织及正常脑组织中DEK mRNA的表达水平。结果低级别、高级别星形细胞肿瘤组织及正常脑组织中DEK mRNA的表达阳性率分别为85.7%、100.0%、20.0%;DEK mRNA相对含量分别为:低级别肿瘤组织0.71±0.07,高级别肿瘤组织0.84±0.06,正常脑组织0.19±0.02。DEK mRNA在肿瘤组织中的表达水平显著高于正常脑组织(P<0.05)。结论 DEK mRNA的表达与星形细胞肿瘤的发生、发展呈显著正相关,它的过表达是星形细胞肿瘤的重要分子生物学特征。 Objective To explore the relationship between DEK mRNA expression and occurrence and development of astrocytoma, and investi- gate the molecule mechanism of astrocytic tumors occurrence. Methods Reverse transcription polymerase chain reaction ( RT- PCR ) technique was employed to detect the levels of expression of DEK mRNA in astrocytoma tissues and brain tissues. Results The rate of positive expression of DEK mRNA was 85.7% in low grade astrocytoma, 100.0% in high grade astrocytoma and 20.0% in brain tissues. The relative coutent of DEK mRN A was 0.71 ±0.07 in low astrocytoma, 0.84±0.06 in high astrocytoma, 0. 19±0.02 in normal brain tissues. The expression of DEK in astrocytoma was significandy higher than brain tissues ( P 〈 0.05 ). Conclusion The expression of DEK is positively correlated with the occurrence and develop- ment of astrocytic tumors. Over-expression of DEK is an important molecule biologic characteristic of astrocytic tumors.
