小麦胚芽抗肿瘤活性蛋白的分离纯化及抗肿瘤活性研究 被引量:3

摘要 脱脂小麦胚芽水溶性提取物经过硫酸铵分级盐析、DEAE-Sepharose Fast Flow离子交换柱层析、Superdex200凝胶柱层析,分离得到抗肿瘤活性蛋白WGWSP11。WGWSP11经高效液相凝胶色谱鉴定显示为单一对称峰,经还原、非还原的SDS-PAGE分析均为单一蛋白染色条带,其相对分子质量约为41 kDa。体外抗肿瘤实验结果显示,WGWSP11对人乳腺癌细胞株MDA-MB231、人胃癌细胞株SGC-7901和人结肠癌细胞株HT-29的细胞存活率均有不同程度的影响,其中对MDA-MB231的影响最为显著,且对正常细胞人胚肾细胞HEK-293的毒性作用很小。这些结果可以为进一步开发小麦胚芽抗肿瘤功能性食品或药物提供一定的思路。 The anti - tumor activity protein of WGWSP11 was separated from water - soluble extract of defatted wheat germ by fractional precipitation with ammonium sulfate, DEAE . Sepharose Fast Flow ion - exchange chromatography and Superdex200 gel filtration chromatography. The WGWSP11 was iden- tified by gel high performance liquid chromatography and the resu]t showed that there was a single sym- metrical peak, and the analysis of reducing and non reducing SDS - PAGE showed that there was a single protein band and the relative molecular weight of WGWSP11 was approximately 41 kDa. The anti - tumor experiments in vitro indicated that WGWSP11 had effects on tlae cell viability of human breast cancer cell line MDA- MB231, human gastric cancer cell line SGC -79D1 and human colon cancer cell line HT- 29, especially the effect on MDA - MB231 was the strongest, and its cytotoxicity on human embryonic kidney cell HEK -293 was very weak. These results could provide some ideas to develop anti - tumor functional foods or drugs from wheat germ.
