摘要
在工业化制造过程中,对微生物总数的监测至关重要,尤其是在食品、药品、电子和化妆品等产业。基于萤火虫荧光素酶的ATP生物发光法是一种快速、简便的方法。尽管在上述领域中有部分认可,但由于该法存在对环境的敏感性、微生物的检测限偏高、缺乏检测结果 RLU的标准等局限性,限制其更为广泛的应用。该研究综合分析了该方法的原理、检测步骤、存在的问题,讨论了通过ATP提取方法、提高荧光素酶活性稳定性、引入扩增体系的研究作为提高方法的灵敏度的切入点,并综述了国内外在此方面的研究进展,为研制更稳定、更精确的生物发光方法提供思路。
Total number of microorganisms monitoring in the process of manufacturing industrial is critical, especially in industries such as food, medicine, electronics and cosmetics. ATP bioluminescence of firefly luciferase-based method is a fast and easy-operated method. Although ap- plied in some of these areas, there are still many limitations, such as high sensitivity to the environment, high detection limitations of microbial and lack of detection criteria of RLU. The principle and testing procedure, limitation problems were analyzed. The sensitivity improvement was discussed from extracting method of ATP, improvement of fluorescein enzyme stability and introducing amplification system as breakthroughs points, and the progress at home and abroad was summarized, in order to provide ideas for the further development of a more stable and more precise methods.
