ERCC1在非小细胞肺癌患者中血液和肿瘤组织中表达的相关性研究

摘要 目的探讨ERCC1在非小细胞肺癌患者中血液和肺癌组织中表达的相关性,以及检测外周血ERCC1的表达水平来预测铂类药物的化疗是否存在耐药的可能性。方法收集42例非小细胞肺癌患者的血液和病理组织,分别利用逆转录定量聚合酶链式反应(RT-qPCR)来检测外周血ERCC1表达水平,免疫组织化学方法来检测病理组织切片的ERCC1表达水平。结果 42例肺癌组织和血液组织中ERCC1表达水平上并没有明显的相关性(P>0.05)。42例肺癌组织和血液组织中ERCC1的表达有统计学意义(P<0.05)。肺鳞癌与肺腺癌组间比较差异具有统计学意义(P=0.038)。结论 ERCC1表达量在外周血和肺癌组织中表达水平有明显的变化。肿瘤组织和血液中的ERCC1的表达无明显相关性。无法应用检测外周血ERCC1的表达水平来预测是否对铂类化疗药物耐药。 Objective To detect the correlation of expression levels of excision repair cross complement group 1 (ERCC1) gene mRNA in non - small cell lung cancer and study their clinical significance. Methods Using RT - qPCR we determined ERCC1 expression levels in blood and using immunohistochemistry we detected the expression of ERCC1 in tumor tissue from the NSCLC patients. Results Target gene expression among different tissues has significant differ- ences (P 〈 0.05). Its expression in pulmonary adenocarcinoma was higher than that in squamous carcinoma (P = 0. 038). A statistically significant correlation (P 〉 0. 05) was not found between target gene mRNA expression in tumor tissue and blood. Conclusion The nonexistence of the correlation of gene expression of ERCC1 in tumor tissue and blood from NSCLC patients should not allow using RT - qPCR to detect the ERCC1 expression in blood then to predict the platinum drug resistance of the patients.
