小动物实验血沉快速测定微量毛细管法的建立与评价 被引量:1

摘要 目的建立一个操作简单、检测快速和结果准确可靠的可广泛用于小动物实验研究的血沉(erythrocyte sedimentation rate,ESR)测定方法。方法以大鼠佐剂型关节炎模型(adjuvant-induced arthritis,AIA)为研究对象,对应用微量毛细管倾斜法进行血沉测定的两个主要变量倾斜角度和测定时间进行优选。结果微量毛细管倾斜角度对血沉的升高影响明显,血沉值由小到大顺序依次为90°<45°<30°。尽管测定时间由15 min延长到30 min再到60 min,实验结果显示测定时间对血沉值影响不明显。与标准魏氏法(Westegren)平行比较,微量毛细管倾斜法测得的血沉值组内离散度更小,组间差异更显著。结论本研究建立的微量毛细管倾斜法,倾斜角度45°,测定时间15 min,可广泛用于小动物实验研究的血沉测定。 Aim To establish method for determination of a simple, rapid, and accurate erythrocyte sedimentation rate (ESR) by using capillary tubes in in vivo experimental models. Methods Adjuvant-induced arthritis (AIA) was established in male Sprague-Dawley (SD) rats, and blood specimens were col- lected from the veins of the tails of AIA and control rats for deter- mination of ESR using the Westegren or capillary tubes. Differ- ent angles of incline of the capillary tubes and durations of ESR determination were screened. Results The angle of incline of the capillary tubes had a significant influence on ESR, with the ESR levels in the order of 90°〈 45° 〈 30°. The duration for ESRdetermination played a relatively minor role in ESR. ESR meas- ured using the capillary tubes held obliquely at an angle of 45 ° for a duration of 15 min was less variable and much more marked than that obtained from the standard Westegren method. Conclu- sion A well-characterized method for determination of ESR u- sing the capillary tubes has been successfully developed, which could be cost-effectively and efficiently used in experimental models of rodents.
