摘要
目的:建立白及药材化学成分的HPLC指纹图谱,为白及药材的质量控制提供科学依据。方法:采用HPLC法,以Waters SunFire C_(18)(250 mm×4.6 mm,5μm)为色谱柱,Diamonsil C_(18)保护柱,以乙腈和水为流动相,梯度洗脱,流速为1.0 ml·min^(-1),检测波长为270 nm,柱温为30℃。结果:15批白及药材化学成分特征图谱共检出9个分离度良好的共有峰。白及药材9个特征峰进行聚类分析,白及药材产地与特征图谱相似度差异不大。样品中参照物1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯(militarine)的含量有明显差别。结论:该法简便、快速,可作为白及的质量控制的有力手段。
Objective: To establish the HPLC fingerprints of chemical constituents in Bletilla striata to provide the basis for the quality control of Bletilla striata. Method: The chromatographic separation was performed on a Waters SunFire Cls (250 mm x 4.6 mm, 5 txm) column with a Diamonsil Cls guard column, and the mobile phase consisted of acetonitrile-water with gradient elution. The flow rate was 1.0 ml ~ min-~ , the detection wavelength was 270 nm, and the column temperature was 30~C. Result: There were 9 common peaks in the HPLC fingerprints of 15 batches ofBletilla striata, and the 9 characteristic peaks showed little change in Bletilla striata from different habitats. There was significant difference in the militarine content among the samples. Conclusion: The HPLC fingerprints method is simple, rapid and feasible in the quality control of Bletilla striata.
