鲜蟾皮外敷促进糖尿病大鼠皮肤溃疡愈合的实验研究 被引量:5

摘要 目的:研究鲜蟾皮外敷对糖尿病大鼠皮肤溃疡的治疗作用及作用机制。方法:清洁级雄性SD大鼠42只,分为3组:正常对照组(12只)、糖尿病皮肤溃疡模型组(15只)、糖尿病皮肤溃疡治疗组(15只)。建立缺损性皮肤溃疡大鼠模型,鲜蟾皮外敷溃疡部位,观察溃疡愈合情况;大体病理观察第2、3周末溃疡组织结构,免疫组化观察血管内皮生长因子(VEGF)的表达,酶联免疫反应技术测定组织中VEGF含量。结果:模型组2周溃疡开始缩小,3周结痂,与对照组相比VEGF表达降低(15.6±2.2μg/g vs.48.3±4.2μg/g,P<0.05);与模型组相比,治疗组2周VEGF表达增加(40.7±4.5μg/g vs.15.6±2.2μg/g,P<0.05),持续到3周(19.5±3.2μg/g vs.10.9±2.7μg/g,P<0.05),创面愈合周期提前。结论:鲜蟾皮外敷可增加VEGF表达,促进糖尿病大鼠皮肤溃疡愈合。 Objective: To study the effect of the treatment of skin ulcers in diabetic rats by external application of fresh toad skin and its possible mechanism of action. Methods: 42 male SD rats were randomly divided into three groups:the normal control group (12 rats), the diabetic skin ulcers model group (15 rats), and the diabetic skin ulcers treatment group (15 rats). A model of skin ulcers in rats was created, then fresh toad skin was applied on these ulcer sites, and how the ulcers got healed was observed. Pathologically, the wounded tissue structures around the ulcers were observed at the 2nd and 3rd weekends, and immunohistochemically, the expression of the wounded tissue vascular endothelial growth factor (VEGF) was observed. Results: Ulcers began to shrink after 2 weeks and scab after 3 weeks in the diabetic model group. By contrast, in the diabetic treatment group, the expression of VEGF was increased (15.6±2.2 μg/g vs. 48.3±4.2 μg/g, P〈0.05) at the end of 2 weeks and sustained to the end of 3 weeks (19.5±3.2μg/g vs. 10.9±2.7μg/g, P〈0.05), and meanwhile the wound healing cycle was shortened. Conclusions:External application of fresh toad skin can increase the expression of the wounded tissues VEGF and promote the healing of skin ulcers in diabetic rats.
