一株硫氧化菌的筛选及其生理生化特性 被引量:1

摘要 通过硫氧化菌的专性培养基筛选法,成功从漳州开发区污水处理厂脱水后的污泥中筛选分离出一株化能自养型硫氧化菌SS 1,对其的形态特征和生理生化性质进行考察鉴定,并研究该株硫氧化菌的金属耐受性及其对硫底物硫代硫酸盐和单质硫的利用性。结果表明:菌株SS 1为革兰氏阴性菌,短杆状,不具有运动性,可以把硝酸盐还原成亚硝酸盐,并且可再进一步还原成氨氮。通过重金属耐受性实验表明,该菌株可耐受浓度为2 mmol/L的Ni、Zn、Co和Cu重金属,对重金属的耐性能力顺序为:Ni2+>Zn2+>Co2+>Cu2+,对重金属Ni的最大耐受性为3mmol/L。菌株SS 1对单质硫的利用率明显高于对硫代硫酸钠的利用,表明其嗜硫效应明显。 An autotrophic rod sulfur-oxidizing strain SS 1 which has capacity of oxidation sulfide was isolated from post- dewatering sludge obtained from sewage treatment plant iu development zone of Zhangzhou city by sulfur-oxidizing bacterium of special medium. Its morphological characteristics and physiology-biochemical properties were observed and identified, besides, heavy metal tolerance and availability of sulfur-substrate (about element sulfitr or thiosulfate) were investigated. The main results were as follow: it was Gram negative and rod-shaped,but immotili/y; Nitrates and nitrites were hoth reduced to ammonia nitrogen by strain SS 1 ; It was tolerable when metal ions (about Ni, Zn, Co and Cu) attained 2 mmol/L; And the tolerance of strain SS 1 for four kinds of heavy metals was in this order: Ni~'+ 〉 Zn^-+ 〉 Co2. 〉 Cu2~ ; It exhibited high minimal inhibitory concentration value (about 3 mmol/L Ni) for Ni2* The strain SS I ' s availability of elemental sulfur was obviously higher than the utilization of sodium thiosulfate, it showed thiophilic of strain SS.
