p53结合蛋白对细胞周期阻滞与细胞凋亡调控的研究进展 被引量:2

摘要 p53基因是人体内重要的肿瘤抑制基因,位于人类染色体17p13.1,含有11个外显子,编码的野生型p53蛋白由393个氨基酸残基组成,包含N.末端的转录激活结构域、生长抑制结构域、序列特异的DNA结合结构域(DNAbindingdomain,DBD)、核定位信号(nuclearlocalizationsignal,NLS)、四聚体化结构域和c.末端非专一DNA调节结构域。p53在监视细胞基因组损伤及维持基因组的稳定性中发挥重要作用。当紫外线、电离辐射、氧化应激及癌基因表达等刺激因素引起细胞内DNA损伤时,p53蛋白迅速聚集活化,并作为序列特异性转录因子对一系列靶基因进行调控。
