摘要
目的探讨爪蟾抗菌肽(magainin)的人工合成及对金黄色葡萄球菌细胞膜的作用。方法利用氨基酸固相合成的方法合成爪蟾抗菌肽,高效液相色谱法进行纯化,质谱鉴定后,得到的爪蟾抗菌肽纯度在99%以上。将爪蟾抗菌肽作用于引起感染性心内膜炎的金黄色葡萄球菌ATCC25923,以PBS缓冲液作为阴性对照,TritonX-100作为阳性对照。结果爪蟾抗菌肽作用于引起心内膜炎的金黄色葡萄球菌后,ATCC25923的630 nm值从0.166下降到0.138,负染后透射电镜观察,发现细菌表面存在破损成小孔的地方,细胞膜界限模糊不清,甚至细胞膜完全溶解。爪蟾抗菌肽通过破坏细菌细胞膜而导致细菌的死亡。结论爪蟾抗菌肽能够提高细菌细胞膜的通透性。
Objective To explore the artificially synthesized method of magainin and its effect on cell mem- brane of Staphylococcus aureus. Methods Magainin was synthesized by amine acid solid-phase synthesis method, and was purified with high efficiency liquid chromatography. Purity of magainin reached above 99% on the basis of mass spectrometry. Staphylococcus aureus ATCC25923 which induce infective endoearditis was cultured and inter- fered with magainin, with PBS buffer as negative control, TritonX-100 as positive control. Results After interfered with magainin, ~630 nm value of Staphylococcus aureus ATCC25923 decreased from 0. 166 to 0. 138. Under nega- tive staining TEM, bacterial surface with hole-like damage was found, and the cell membrane boundaries blurred, or even completely dissolved at the hole-like damage. Conclusion Artificially synthesized magainin' can improve the oermeabilitv of the bacterial cell wall.
