血小板胶-胶原复合膜修复大鼠牙周组织缺损的疗效观察 被引量:1

The effect of APG-Co membrane in repairing periodontal tissue defects of rats

摘要 目的探讨血小板胶(APG)-胶原(Co)复合膜对大鼠牙周组织缺损的修复作用。方法将Co分别裁成5 mm×2 mm及7 mm×4 mm大小,抽取大鼠全血10 ml制作富血小板血浆(PRP),再将5 mm×2 mm Co浸润于PRP中制备APG-Co复合膜。将16只成年雄性Wistar大鼠随机平均分为实验组和对照组,分别制作大鼠下颌牙周骨缺损模型(缺损大小为5 mm×2 mm,磨除牙根面牙骨质)。实验组置APG-Co复合膜于缺损内,并于缺损表面覆盖7 mm×4 mm的Co。对照组仅用7 mm×4 mm Co覆盖缺损表面。术后4、6周每组分别处死4只,HE染色观察牙周组织缺损修复效果,Image-Pro Plus对术后6周HE染色结果进行分析,计算出新生牙周骨面积,应用SPSS 13.0软件对新生牙周骨面积进行统计学分析。结果实验组新生牙槽骨和牙骨质样组织的生成量和成熟度均优于对照组,术后6周实验组新生牙周骨面积显著高于对照组[(56 533.04±2 673.61)vs(23 006.05±2 198.10)μm2,P<0.05]。结论 APG-Co复合膜有显著的促进牙周骨组织缺损修复的作用。 Objective To investigate the effect of APG (autologous platelet gel)-Co (collagen) membrane on repairing periodontal tissue defects in rats. Methods The Co was cut into 5 mm x 2 mm and 7 mm × 4 mm sizes respectively, and 10 ml whole blood of rat was extracted to prepare platelet-rich plasma ( PRP), then the 5 mm × 2 mm Co was infiltrated into PRP to make APG-Co membranes. 16 Wistar rats were randomly divided into experimental group and control group ( n = 8 each ). The mandible alveolar periodontal bone defect model ( defect size: 5 mm× 2 mm, root cementums were struck off) was made. In experimental group, APG-Co membrane was put into periodontal bone defect, and then the 7 mm× 4 mm Co covered the surface of defect. In control group, the 7 mm × 4 mm Co was used only to cover the surface of defect. Four rats of each group were killed respectively at the 4th week and the 6th week after establishing models. The repairing effect of periodontal tissues defect was observed by HE dyeing, and the result was analyzed by Image-Pro Plus to calculate the area of new born bones 6 weeks later. Then the areas of new born bones were conducted by SPSS 13.0 software to do statistical analysis. Results The quantity and degree of maturation of newly formed alveolar bone and cementoid tissues in experimental group were all better than those in control group. The area of newly formed periodontal bone 6 weeks later in experimental group was significantly higher than that in control group[ (56 533.04 ± 2 673.61 ) μm^2 vs ( 23 006.05 ± 2 198.10) μm^2, P 〈 0. 05 ]. Conclusions APG-Co membrane plays obvious role in promoting the repair of periodontal bony tissue.
