孕鼠锌摄入与外界不良因素共同作用对子代腭裂发生的影响 被引量:2

摘要 目的:探讨孕鼠缺锌等不良因素对子代腭裂畸形发生的影响和锌在腭发育中的作用。方法:将42只C57BL/6 E0孕鼠按不同处理因素分为对照组、高温常锌组(A组)、高温高锌组(B组),高温低锌组(C组),内毒素常锌组(a组),内毒素高锌组(b组)、内毒素低锌组(c组)(n=6),高温为42.5℃水浴,9 min/次,1次/6 h。常锌饲料含锌30 mg/kg,高锌饲料100 mg/kg,低锌饲料5 mg/kg。内毒素为大肠杆菌脂多糖0.5μg/g,皮下注射。于胚胎18 d处死孕鼠,观察胎鼠腭裂发生情况。结果:对照组和实验A、B、C、a、b、c组胎鼠腭裂发生率分别为3.03%、24.13%、3.45%、36.67%、26.67%、5.71%、38.71%。对照组与B或b组比较,P>0.05,与其余试验组比较,P<0.05;实验B组与A或C组比较,P<0.05,实验b组与a或c组比较,P<0.05。结论:母体血清锌含量与子代腭裂的发生有密切关系,高锌可以减少子代在不良刺激因素下的腭裂发生,但低锌与高温和内毒素不良因素的致子代腭裂畸形作用并无叠加。 Objective:To study the effect of Zinc intaking and environmental factors on the incidence of fetus cleft palate of mice.Methods:42 fertilized C57 BL/6 mice were divided into seven groups (n =6) at E0:control group,normal zinc food + high temperature group (A),high zinc food + high temperature group (B),low zinc food + high temperature group (C),normal zinc food + endotoxin group (a),high zinc food + endotoxin group (b),low zinc food + endotoxin group (c).Normal zinc food contained 30 mg/kg of zinc,high zinc food 100 mg/kg and low zinc food 5 mg/kg.High temperature was 42.5 ℃ water bath treatment for 9 min once every 6 h.Colibacillus lipopolysaccharide at 0.5 μg/g was given by subcutaneous injection as the endotoxin treatment.The mice were killed at E18.5 and fetus were collected.The mouse embryonic cleft palate was observed under stereo-microscope.Results:The incidence of cleft palate was 3.03%,24.13%,3.45%,36.67%,26.67%,5.71% and 38.71% in the group of control,A,B,C,a,b and c respectively,control vs A,C,a or c,P < 0.05 ; B vs A or C,b vs a or c,P < 0.05.Conclusion:The high content of the serum zinc in pregnant female mice can decrease the risk of cleft palate in fetus when the female were affected by adverse environmental factors during pregnancy.The low content of the serum zinc in the pregnant female mice and high temperature or endotoxin have no additive effect on the risk of cleft palate in fetus.
