摘要
目的通过在体外成熟(IVM)培养基中加入褪黑素,探索外源性褪黑素在人类未成熟卵母细胞IVM中的作用,以期优化人类未成熟卵母细胞IVM的培养体系。方法在人类未成熟卵母细胞IVM培养体系中添加不同浓度的褪黑素,以排出第一极体作为判断其核成熟的标准,比较不同浓度下的核成熟率。结果与对照组(褪黑素0 mol/L)相比,外源性褪黑素在10^(-11)mol/L、10^(-9)mol/L和10^(-7)mol/L时人类未成熟卵母细胞的核成熟率有提高,但只有褪黑素在10^(-9)mol/L时人类未成熟卵母细胞的核成熟率呈显著性提高(63.6%vs.40.9%,87.7 vs.54.2%,68.2%vs.45.5%,90.4 vs.69.4%)(P<0.05);当外源性褪黑素浓度增加到10^(-5)mml/L和10^(-3)mol/L时,人类未成熟卵母细胞的核成熟率显示稍低于对照组,差异无统计学意义(P>0.05),但与10^(-11)mol/L、10^(-9)mol/L和10^(-7)mol/L组相比则呈显著性降低(P<0.05)。结论外源性褪黑素在适当浓度时可以促进人类未成熟卵母细胞的核成熟,而在浓度超出一定范围时,则可能对卵母细胞的核成熟造成负面影响。
Objective: To investigate the effect of melatonin on in vitro maturation of human oocytes. Methods: Human immature oocytes were cultured in in vitro maturation medium supplemented with different concentration of melatonin. The rate of nuclear maturation was evaluated by the presence of first polar body. Results: Nuclear maturation rate of human oocyte was improved in 10^11 mot/L,10^-9 mol/L and 10^7 mol/L melatonin treatment groups. And nuclear maturation rate was improved significantly when melatonin was supplemented in 10^-9 mol/L (P〈0.05). Nuclear maturation rate of human oocyte was slightly decreased in 10^-5 mol/L or 10^-3 mol/L melatonin treatment groups compared with the control group,which was significantly lower than that in 10^-11mol/L, 10^-9 mol/L and 10^-7 mol/L melatonin treatment groups(P〈0.05). Conclusions: Exogenous melatonin at a proper concentration has beneficial effect on nuclear maturation of human immature oocytes. And it has adverse effect on nuclear maturation of human immature oocytes to add melatonin at high concentration.
