大豆三羟基异黄酮对大鼠急性胰腺炎的影响

摘要 目的探讨大豆提取物三羟基异黄酮(Genistein)对大鼠急性胰腺炎的影响。方法 45只SD大鼠随机分为3组:3组均采用精氨酸腹腔注射诱导建立急性胰腺炎模型,对照组造模后经口胃内灌注大豆蛋白;模型组经口胃内灌注生理盐水;实验组经口胃内灌注三羟基异黄酮。观察大鼠72 h内生存与死亡情况;检测存活大鼠24 h、48 h、72 h血清淀粉酶含量,计算胰腺组织病理指数评分。结果大豆蛋白组6 h死亡率高达53.3%;三羟基异黄酮组6 h累计死亡率仅为6.7%,至24 h达死亡高峰(33.3%),较大豆蛋白组明显延后,两组死亡率比较差异有统计学意义(P<0.01)。三羟基异黄酮组存活大鼠24 h、48 h、72 h血清淀粉酶活性显著低于大豆蛋白组,差异有统计学意义(1830 U/L±325 U/L vs 2667 U/L±262 U/L,P<0.01;1744 U/L±321 U/L vs 2935 U/L±301 U/L,P<0.01;1319 U/L±338 U/L vs 2725 U/L±235 U/L,P<0.05);3组胰腺炎症病理指数差异有统计学意义。结论经胃灌注三羟基异黄酮可以延缓急性胰腺炎大鼠死亡高峰,提高大鼠生存率,抑制血清胰腺淀粉酶活性,可能通过降低胰腺炎症反应而发挥重要作用。 Objective To investigate the effect of Genistein derived from soybean on rats with acute pancreatitis.Methods Acute pancreatitis model was induced by intraperitoneal injection with arginine (2-amino-5-guanidinovaleric acid).Then 45 SD rats were randomly divided into 3 groups:control group was intragastric administration of soybean protein (100 g/kg) three times every day,model group was given the same dose of normal saline,and Genistein group was given 5,7,4'-triatomic isoflavone (100 mg/kg).The rat survival and death within 72 h of situation were observed,serum amylase levels at 24 h,48 h,72 h were detected,and pancreatic tissue pathological index score was calculated.Results The mortality rate of contorl group at 6 h was the highest (53.3%),while that of Genistein group was only 6.7%.The peak mortality of Genistein group was at 24 h (33.3%),and there was significant difference compared with control group (P <0.01).The serum amylase activity at 24 h,48 h,72 h in Genistein group was significantly lower than that in control group (1830 U/L ±325 U/L vs 2667 U/L ±262 U/L,P <0.01 ; 1744 U/L ±321 U/Lvs2935 U/L±301 U/L,P<0.01; 1319 U/L±338 U/Lvs2725 U/L± 235 U/L,P<0.05).Further,there was significantly different in pancreatic tissue pathological index score among three groups.Conclusion Intragastric administration of Genistein can delay the peak of death of rats with acute pancreatitis,improve the survival rate of rats,and inhibit the serum pancreatic amylase activity,probably by reducing pancreatic inflammation.
